Bontac Bio-Engineering (Shenzhen) Co., Ltd. (hereafter referred to as BONTAC) is a high-tech enterprise established in July 2012. BONTAC integrates R&D, production and sales, with enzyme catalysis technology as the core and coenzyme and natural products as main products. There are six major series of products in BONTAC, involving coenzymes, natural products, sugar substitutes, cosmetics, dietary supplements and medical intermediates.
As the leader of the global NMN industry, BONTAC has the first whole-enzyme catalysis technology in China. Our coenzyme products are widely used in health industry, medical & beauty, green agriculture, biomedicine and other fields. BONTAC adheres to independent innovation, with more than 170 invention patents. Different from the traditional chemical synthesis and fermentation industry, BONTAC has advantages of green low-carbon and high-value-added biosynthesis technology. What’s more, BONTAC has established the first coenzyme engineering technology research center at the provincial level in China which also is the sole in Guangdong Province.
In the future, BONTAC will focus on its advantages of green, low-carbon and high-value-added biosynthesis technology, and build ecological relationship with academia as well as upstream/downstream partners, continuously leading the synthetic biological industry and creating a better life for human beings.
The preparation methods utilized by manufacturers worldwide of NAD powder are mainly divided into chemical synthesis method and biocatalytic method, among which biocatalytic method includes biological fermentation method and enzyme catalysis method. Enzyme catalysis method has gradually become the mainstream direction because of its advantages of green, environmental protection and pollution-free. And then the purity of NAD powder will reach 99% after the procedure of further purifying. But some NAD manufacturers tend to choose the multiple methods to develop the producing of NAD.
Molecules that can be taken in supplement form to increase NAD levels in the body are referred to by some as “NAD boosters.” Studies conducted over the past six decades suggest that the following are some of the many benefits associated with taking an NAD supplement:
Can Help Restore Mitochondrial Function
Helps Repair Blood Vessels —A 2018 mice study found that supplementation could aid in repair and growth of aged blood vessels. There’s also some evidence it can help manage heart disease risk factors like high blood pressure and high cholesterol.
May Improve Muscle Function — One animal study conducted in 2016 found that degenerative muscles had improved muscle function when supplemented with NAD+ precursors.
Potentially Helps Repair Cells and Damaged DNA — Some studies have found evidence that NAD+ precursor supplementation leads to an increase in DNA damage repair. NAD+ is broken down into two component parts, nicotinamide and ADP-ribose, which combine with proteins to repair cells.
May Help Improve Cognitive Function — Several studies conducted on mice have found that mice treated with NAD+ precursors experienced improvements in cognitive function, learning and memory. Findings have led researchers to believe that NAD supplement may help protect against cognitive decline/Alzheimer’s disease.
May Help Prevent Age-Related Weight Gain — A 2012 study showed that when mice fed a high-fat diet were given an NAD supplement, they gained 60 percent less weight than they did on the same diets without the supplement. One reason this may be true is that nicotinamide adenine dinucleotide helps regulate production of stress- and appetite-related hormones, thanks to its effects on circadian rhythms.
Precursors are molecules used in chemical reactions inside the body to create other compounds. There are a number of precursors of NAD+ that result in higher levels when you consume enough of them.

1.Prevention and treatment of viral-induced inflammatory storms
Scientists have found after extensive research that neo-coronavirus has a mechanism similar to SARS virus to activate inflammatory vesicles NLRP3. and the activation of NLRP3 produces more inflammatory factors, generating excessive inflammation and thus triggering a deadly cytokine storm. This problem can be well addressed by NAD+, which inhibits the activity of NF-κB inflammatory pathway and NLRP3 inflammasome by increasing the activity of sirtuins (SIRT1, SIRT2 and SIRT3), thus preventing cytokine storm caused by excessive inflammation. Therefore, Sinclair and other scientists believe that increasing the concentration of NAD+ may play an important role in the prevention and treatment of neocoronavirus and other viral infections.
2.Restoration of virus-induced metabolic disorders
NAD+ is an essential coenzyme for many cellular energy metabolic pathways, present in every cell of the body, involved in thousands of reactions, and an important player in maintaining cellular viability. In the COVID-19 infection model, NAD+ and NMN supplementation was found to be effective in alleviating cell death and protecting the lung.
First, inspect the factory. After some screening, NAD manufactures that directly face consumers pay more attention to brand building. Therefore, for a good brand, quality is the most important thing, and the first thing to control the quality of raw materials is to inspect the factory. Bontac company actually manufacturing NAD powder of high quality with the caterias of SGS. Secondly, the purity is tested. Purity is one of the most important parameters of NAD powder. If high purity NAD cannot be guaranteed, the remaining substances are likely to exceed the relevant standards. As the attached certificates demonstrates that the NAD powder produced by Bontac reach the purity of 99.9%. Finally, a professional test spectrum is needed to prove it. Common methods for determining the structure of an organic compound include Nuclear Magnetic Resonance Spectroscopy (NMR) and high-resolution mass spectrometry (HRMS). Usually through the analysis of these two spectra, the structure of the compound can be preliminarily determined.
In the international market, NAD+ food supplements are in production in the United States, the Australian Food and Drug Administration has approved NAD+ products for sale, and Japanese NAD+ manufactures are devoted to extend the market share in China. Li Ka-shing invested HK$200 million in the US NAD manufacturer, whose NAD+ products are being sold by Watsons in Hong Kong! Then Mclane, a company owned by Warren Buffett also joined the NAD market.
Introduction Wound healing is a sophisticated process responding to tissue damage, which is associated with numbers of interaction of various cell types, cytokines, growth factors, and other molecules. Strikingly, increasing the nicotinamide adenine dinucleotide (NAD) pool by nicotinamide riboside (NR) can accelerate wound healing and macrophage migration, which is partially achieved through PGE2 synthesis and signaling as well as the function of the NAD+-dependent sirtuin, SIRT3. Regulatory effects of NR on the expression of M1 macrophage markers in human MDMs. NR could modulate the expression levels of canonical M1 (inflammatory phenotype) and M2 (reparative phenotype) cell surface markers during macrophage polarization. With a great detail, a significant downregulation in CD64 and a obvious upregulation of CD197/CCR7 are viewed in the polarized M1 cells incubated with NR. Furthermore, NR increases CD197/CCR7-mediated M1 macrophage migration. The significance of chemotaxis mediator PGE2 in NR-regulated macrophage migration NR-mediated upregulation of macrophage migration through CCL19/CCR7 is dependent on the synthesis of PGE2, an inflammatory lipid mediator in the eicosanoid family. Concretely, NR administration increases the PGE2 level in cultured human monocytes, MDMs, and human serum. In addition, NR-mediated increases in CCR7 expression and CCL19-induced migration are attenuated by PGE2 synthesis blockers. NR/SIRT3/migration axis in human M1 MDMs NR facilitates collective cell migration at a SIRT3-dependent manner in human M1 MDMs during wound healing. Simply put, the degree of wound healing is compared on Day 0 and Day 2 in vehicle- or NR-treated human M1 MDMs. It is found that NR increases the relative degree of migration (relative wound healing) and the rate of wound confluence in the presence of CCL19. Besides, the relative degree of wound density (migration) is blunted by SIRT3 knockdown, while being enhanced by SIRT3 overexpression. Application prospect of NR in wound healing Chronic diabetes is often accompanied with poor wound healing. For instance, diabetic foot ulcers, one of the chief cause of amputations, affect 15% of people with diabetes. Given that NR can drive the macrophage migration to boost chronic wound healing, it may have a broad application prospect in treating the wounds including but not limited to diabetic patients. Conclusion In human macrophages, NR induces surface expression of the chemotaxis CD197/CCR7 receptor and levels of its lipid mediator PGE2 via upregulation of cyclooxygenase 2 and functionally increases macrophage migration and wound healing in a SIRT3-dependent manner. Introduction Ginsenoside Rg3 (Rg3) holds great promise as a potential drug for preventing scar formation, but it is often difficult for it to work alone due to its poor water solubility and low bioavailability. Herein, a novel Rg3-loaded dermal extracellular matrix (DECM) hydrogel scaffolds are prepared via 3D printing and nanoloading technologies, which improves the biosafety profile of Rg3 to a large extent, maintains a consistent local drug concentration for an extended duration and prolongs therapeutic effects, realizing the final goal of scar-free healing. The merits of 3D-printed DECM hydrogel scaffolds loaded with Rg3 3D-printed DECM hydrogel scaffolds have good supportability and stability. They could be folded into different shapes without obvious deformation or breakage, closely resembling the properties of natural skin. The continuous porous structure of DECM-2MSN scaffolds enables the faster transport of nutrients, oxygen, water, and metabolic wastes within the platform, which facilitates the wound healing. The uniqueness of 3D-printed DECM hydrogel scaffolds loaded with Rg3 Through decellularization, about 98.57% of dsDNA is removed from DECM, leaving few cellular remnants while preserving collagen in DECM, which simulates the natural extracellular matrix microenvironment for skin defect healing to the greatest extent, reducing the risk of immune rejection. Hydrogel exhibits excellent biocompatibility and solid-like rheological properties. Furthermore, mesoporous silica nanoparticles (MSNs) are introduced into the system to encase the Rg3 to control its release rate and enhance its bioavailability. The role of Rg3-loaded hydrogel scaffolds in wound healing The wound in DECM-2MSNs/Rg3 group has completely healed, exhibiting smooth and uniform regenerated epithelium with clear boundaries between the epidermis and dermis. During the wound healing process, Rg3 can suppress the excessive inflammation, hinder the formation of blood vessels, and prevent excessive and disordered proliferation of granulation tissue at the wound site, as manifested by the low expression of CD31, VEGF and TGF-β levels in DECM-2MSNs/Rg3 group. In the later stages of wound healing, Rg3 could repress collagen accumulation, thereby impeding scarring, without affecting the normal healing of the wound. Conclusion Rg3-loaded hydrogel scaffold can inhibit wound inflammation and collagen accumulation to repress scar formation. By integrating 3D bioprinting and nanoloading technologies to prepare this innovative wound dressing, the efficacy of Rg3 has been greatly improved, providing a novel therapeutic approach for scar-free wound healing. Introduction Cigarettes containing nicotine (Nic) and sugar-sweetened beverages (SSBs) with high fructose corn syrup (HFCS) have been uncovered to induce hepatic steatosis, which are the risk factors for the development of metabolic diseases including non-alcoholic fatty liver disease (NAFLD). Noteworthily, nicotinamide riboside (NR) can elevate mitochondrial nicotinamide adenine dinucleotide (NAD+) to exert a protect role against hepatic steatosis in NAFLD. About NAFLD NAFLD, which is marked by excessive neutral fat accumulation in the liver, is a frequent chronic liver disease unrelated to alcohol and other well-defined liver-damaging factors, with a global prevalence of 25%. With the epidemic trend of obesity and metabolism-associated syndromes worldwide, NAFLD has been regarded as a type of metabolic stress liver injury with tight association with insulin resistance and genetic predisposition. The association between Nic/Coke and hepatic steatosis in NAFLD Nic plus Coke administration causes hepatic steatosis. In addition, Nic plus Coke also leads to a striking increase in lipid accumulation and mitochondrial swelling as well as loss of the mitochondrial cristae (arrowhead), with a decrease in the amount of endoplasmic reticulum (arrow) and glycogen deposition (asterisk). The preventive role of NR against hepatic steatosis in NAFLD NR leads to a decrease in the calorie intake to reduce hepatic steatosis through the increased of the NAD+ concentration, and triggers the downstream signaling of NAD+, with a reduction in oxidative stress markers and mitochondrial damage. Underpinned mechanism of NR in ameliorating Nic plus Coke-induced hepatic steatosis Nic plus Coke consumption reduces the protein levels of the NAMPT, diminishing NAD+-dependent enzyme Sirt1 and downstream PGC1α. This combined treatment also enhances SREBP1c and ACC activity, increasing intracellular triglyceride deposits and reducing AMPK phosphorylation. The decline in PGC1α can result in the retention of damaged mitochondria with increased oxidative stress, indicated by higher MDA and HO1 levels as well as lower SOD2 level. Importantly, the reduced hepatic NAD+ level is a risk factor to develop NAFLD. NR is a precursor of NAD+ that reduces plasma triglycerides and total cholesterol levels, in turn preventing hepatic lipid accumulation in mice and humans. Sirt1, an NAD+-dependent enzyme, has a protective effect in NAFLD. Sirt1 promotes mitochondrial biogenesis through PGC1α. NR supplementation activates Sirt1 signaling to repress the progression of NAFLD. Conclusion NR supplement has the potential to mitigate oxidative stress and mitochondrial abnormalities, and prevent hepatic steatosis induced by Nic plus Coke, which may be promissory candidates for the management of NAFLD.
